With all the new changes to parent residence visa, can my partner sponsor my father alone (which means a person sponsoring their father-in-law) if he meets the income cut off and other requirements for a single sponsor?

No.

F4.35(a) explicitly states that a parent must be sponsored by either the adult child alone (who meets the family requirements outlined at F4.30.1), or an adult child together with their partner.
